Mitotic inhibitors include-
Etoposide, Vincristine, Vinblastine, Teniposide, Vinorelbine tartarate

Mitotic inhibitors include-

Etoposide, Vincristine, Vinblastine, Teniposide, Vinorelbine tartarate

Adverse Reaction:

Severe myleosuppression.
Nausea, vomiting,
Anorexia, diarrhoea,

Stomatitis, reversible alopecia.
Rarely disturbances of liver function,

Peripheral neuropathy.
CNS effects,

Anaphylactoid reactions.
Hypotension (with I.V.injection).

Contra-Indications:

Hypersensitivity, severe liver dysfunction, pregnancy.

Special precautions:
Caution in handling and preparing solutions for injection.
Skin reaction may occur with accidental exposure.
Use of gloves recommended.
If contact with skin or mucosa occurs, wash affected part with soap and water throughly.
Lactation.

Dosages/ Overdosage Etc:

Indications:

Acute monocytic and myleomonocytic leukemias.

Dosage:
Parentral - 35mg/sqm/day for 4 days to 50mg/sqm/day for 5 days. Courses are repeated at 3 to 4 weks intervals. After recovery from toxicity.

1.Allergies-

Tell your doctor if you are have ever had any unusual or allergic reaction to

etoposide..

2.Pregnancy-

There is a good chance that this medicine will cause birth defects if it is being

used at the time of conception. It is best to use some kind of birth control while

you taking etoposide.

3. Breast-feeding-

Because etoposide may cause serious side effects, breast feeding is not

recommended while you are receiving etoposide

4.Children-

Although this medicine has been used in children, there is no specific information

comparing the use of etoposide in children with use in other age groups.

5.Older adults-

Many medicines have not been studied specifically in older people. Therefore,

it may not be known whether they work exactly the same way they do in

younger adults, or if they cause different side effects or problems in older people.

There is no specific information comparing use of etoposide in the elderly with use

in other age groups

6. Other medicines-

Although certain medicines should not be used together at all, in other cases two

different medicines may be used together even if an interaction might occur.

In such cases your doctor may want to change the dose, or other precautions

may be necessary.

Tell your doctor if you are using any of the following-

Amphotericin by injection

Chloramphenicol

Colchicine

Flucytosine

Gangiciclovir

Interferon

Pilacamycin

Zidovudine

If you have been treated with radiation or cancer medicine - etoposode

may interfere with the effects of these medicines or radiation therapy

on the blood.

7. Other medical problems-

The presence of other medical problems may affect the use of etoposide.

Make sure you tell your doctor if you have any other medical problems

especially-

Chickenpox or Including recent exposre)

Herpes zoster - risk of severe disease affecting other parts of the body

Infection- etoposide can decrease your bodys ability to fight infection.

Kidney disease or

Liver disease effects of etoposide may be increased because of slower

removal from the body.

Pharmacology/ Pharmacokinetics:

Pharmacology:
Etoposides main effect appears to be at the G2 portion of the cell phase.. The predominant
macromolecular effect appears to be DNA synthesis inhibition.

Pharmacokinetics:
The mean oral bioavailability is approx 50%. The disposition of etoposide is a biphasic process
with a distribution half-life of about 1.5 hours.

Interaction with Food:

None reported.

Pregnancy and lactation:

Pregnancy:
Etoposide and tenoposide may cause fetal harm. Decide whether to discontunue the drug or not
based on the relative importance of the drug to the mother.

Lactation:
Not known whether the drug is excreted in breast milk. Decide whether to discontinue nursing or
the drug depending on the importance of the drug to the mother.
