Polythemia vera, chronic granulocytic leukemia in patients with refractory to busulfan

 Alkylating Agents Include-

Bone marrow depression causing leukopenia, thrombocytopenia, anemia, vomiting diarrhea, nausea, abdominal cramps, rash.

Patients with bone marrow depression from x-ray or cytotoxic chemotherapy Special precautions: Perform bone marrow studies prior to treatment and at the time of maximal hematologic response. Perform complete blood counts once or twice daily. Perform liver and kidney function tests

Polythemia vera, chronic granulocytic leukemia in patients with refractory to busulfan

Dosage:

Initially 1mg.kg/day. Larger doses (1.5 to 3mg/kg/day) may be required in patients in patients refractory to other reatment, but do not use as such doses until a dose of 1mg/kg/day has been given for at least 30 days without improvement.

1.Notify physicain if nausea, vomiting, cramps, diarrhea, and rash becomes pronounced.

2.Promptly report fever, sore throat, signs of local infection, easy bruising, bleeding from site or symptoms of anemia

3.Contraceptive measures are recommended during therapy

Pharmacology: Pipobroman is classified as an alkylating agent, but exact mechanism of action is not understood. Pharmacokinetics: Metabolic fate and excretion route are unknown.

Pregnancy:

Advise women of child bearing potential to avoid pregnancy

Lactation:

Excercise caution, and decide whether to discontinue nursing or discontinue the drug,taking into account the importance of the drug to the mother.
