Adverse Reaction:
Abrupt withdrawal of narcotic depression may result in Nausea, vomiting, sweating, tachcardia, increased blood pressure, and tremulousness. In postoperative patients excessive dosage may result in excitement and significant reversal of analgesia, hypotension,pulmonary edema, and ventricular tachycardia and fibrillation. Seizures have been reported infrequently.
Contra-Indications:
Hypersens.
Special precautions:
Hypertension.
Other supportive therapy- mainatin a freeway and provide artificial ventilation, cardiac massage and vasopressor agents, empoly when necessary to counteract acute narcotic overdosage.
Cradiovascular effects- use caution in patients with preexisting cardiac disease or who have received potentially cardiotoxic drugs
Warnings- Drug dependence- administer cautiously in patients who are known or suspected to be physically dependent on opioids, including newborns of mothers with narcotic dependence. Reversal of narcotic efect will precipitate acute abstinence syndrome.
Repeat administration- administer repeated doses as necessary since the duratin of actionof some of the narcotics may exceed that of the narcotic antagonoist.
Respiratory depression- reversal of buprenorphine-induced respiratory depression may be incomplete , if an incomplete response occurs, mechanically assist respiration.
Pregnancy- usecduring pregnancy only when needed.
Lactation- use caution when administering to a nursing woman.
Dosages/ Overdosage Etc:
Indications:
Reversal of narcotic depression.
Dosage:
Give IV,IM or SC. Adults- initial 0.4 to 2mg IV,may repeat IV at 2 or 3 minutes intervals.
Children-
initial- 0.01mg/kg IV.Give subsequent dose of 0.1mg/kg if needed.

Pharmacology:
The mechanism of cation is not clearly understood,evidence suggests that it antagonises the opoid effects by competing fir the same receptor sites.
Pharmacokinetics:
Naloxone is rapidly distributed in the body. It is metabolised in the liver, primarily by glucuronide conjugation. It is excrerted in the urine. The serum half-life in adults ranges from 30 to 80 minutes.
