Tertiary amines - Amitriptyline,Clomipramine,Doxepin,Imipramine,Trimipramine
Secondary amines - Amoxipine, Desipramine, Nortriptyline,Portriptyline
Refer - Amitriptyline
Impairs antihypertensive effect with guanethedine,methyl dopa and methanidine. Potentiates hypertension produced by sympathtomimetics with MAOIs. Reduced plasma levels with barbiturates Raised levels with methylphenedate. Alcohol potentiates sedative effect.

Adverse Reaction:
Agranulocytosis,thrombocytopenia. Drowsiness,orthostatic hypotension and tachycardia(elderly), Headache,peripheral neuropathy tremor,ataxia,seizures, tinnitus, Confusion/delerium(elderly)
Metallic taste,stomatitis, Gastric irritation, Nausea and vomiting, Allergic skin reactions.
Contra-Indications:
Recent M.I.,cardiac arrhythmias,heart block.
Special precautions:
Mania,severe liver disease,narrow angle glaucoma.

Dosage:
Adult- initially 50 -75 mg/day in divided doses. Increase if necessary to 150 mg/day. may be given in divided doses or as a single dose at night
Do not exceed 200mg/day . Elderly - initial 50 -75 mg daily increased gradually Max 100mg daily
