Zidovudine 

Acyclovir and acyclovir , valacyclovir may increase CNS deppression Increased risk of haemotologic toxicity with ganciclovir , valganciclovir, dapsone, doxirubin, vinceristine and vinblastine

Doxirubicin may reduce phosphorylation Fluconazole may increase toxic levels/effects Increased risk of ghepatic decompensation or haemotological toxicites with interferon-alpha and ribavarin.( also increased risk of pancreatitis and lactic acidosis

Methadone may increase effects/levels Increased risk of myalgia, malaise and or fever, maculopapular rash and effects /levels with probenecid Statvudine may decrease antiviral activity

Valproic acid may increase plasma levels AUC increased by 80%

Lamivudine- Increaed risk of hepatic decompensation or haematolgical toxicites with interferon- alpha and ribavarin also increased risk of mitochondrial toxicity, pancreatic and lactic acidosis

Ganciclovir and vaganciclovir may increase effects/toxicity Sulfamethozole/ trimetroprin- may increase AUC and decrease clearance levels ( increasing levels/effects)

HIV infection

Headache, malaise, and fatigue, fever or chills, nausea, diarrhoea, Anorexia, abdominal pain, neuropathy, insomnia and other sleep disturbances

Dizziness, depressive disorders, Nasal signs and symptoms Skin rashes, musculoskeletal pain,myalgia, Anemia, neutropenia, and leucopenia seen at high doses of zidovudine 1200- 1500mg /day seen 4-5 weeks after therapy

Neutrophil count - < 0.75* 10 raised to 9/l Hb levels < 7.5g/dl or 4.65mmol/l lactation

Special precautions

Renal and hepatic impairment, poor bone marrow reserve prior to treatment Risk of opputunistic infection Patients with obese women - with risk factors or active liver disease

Patients with low body weight should avoid combined formulation

Monitor haematalogical parameters -advanced disease 2 weekly for 1st , 3mths, of treatment, Then monthly , early disease 1-3 mthly
