Interaction with Food
Pregnancy and lactation
Alcohol,barbiturates and CNS depressants- trazodone may enhance the CNS depressant response to these agents.
Digoxin- serum levels are increased.
Phenytoin- serum levels are increased
Warfarin- Coadministration with warfarin decreased prothrombin time.The hyperprothrombinemic effect of warfarin may be decreased.
Drug/Lab test interaction: Occassional low white blood cell and neutrophil count has been noted
Triazolopyridine group - Trazadone
Edema, allergic reactions, rash,pruritis. Hypertension, hypotension, shortness of breath, sycope, tachycardia, palpitations, chest pain.
Anger,hostility,nightmares/vivid dreams, Confuson, disorientation, Decreased concentration,dizziness, lightheadedness. GI disorders,anaemia.
Hypersensitivity to trazodone.
Possibility of suicide in severely depressed patients. Prescriptions should be written to the smallest number of tablets. Hypotension and sycope
Avoid concurrent admin with electro-convulsive therapy.
May produce drowsiness,dizziness and blurred vision,
Patients should observe caution while driving or performing tasks which require alertness.
Dosages/ Overdosage Etc:
Dosage:: Adults- Initial dose 150mg/day. This may be increased by 50 mg/day every 3 to 4 days. Maximum dose should not exceed 400 mg/day in divided doses.
Mechanism of antidepressant actions in human not fully understood. Trazadone is not a monoamine oxidase inhibitor,and unlike amphetamine-type drugs,does not stimulate the CNs.
Well absorbed after oral administration without selective localisation in any tissue. Peak plasma levels in approximately 1 hour when taken on an empty stomach or in 2 hours when taken with food.Metabolism: Extensively metabolised in the liver; less than 1 % excreted unchanged in urine and feces. Half-life of 3 to 6 hours(mean 4.4)..

Pregnancy and lactation:
Pregnancy: Trazadone has caused increased fetal resorption and congenital abnormalities in rat and rabbit. No adequate and well controlled studies in pregnant women. Use during pregnancy only when required.
Lactation: Trazodone and its metabolites have been found in the milk of rats,suggesting that the drug may be excreted in breast milk. Excercise caution while administering to a nursing mother.
