Indication:
Prevention of premature ovulation in controlled ovarian stimulation followed by oocyte pick-up and assisted reproductive techniques
Adverse Reaction:
Injection site reactions Ovarian hyperstimulation syndrome Headache, nausea Elevated enzyme e.g. ALT , alkaline phosphatase
Contra-Indications:
Moderate to severe renal or hepatic imapirment Post menopause Pregnancy Lactation Extrinsic pedptide hormone Mannitol hypersensitivity
Special pecautions- Repeated ovarian stimulation oe cycles, Active allergic conditions Do not complete gynaecolgical and endocrinal exam first
Dosages/ Overdosage Etc:
Prevention of premature ovulation in controlled ovarian stimulation followed by oocyte pick-up and assisted reproductive techniques 0.25mg once daily by s.c injection into lower abdomen wall given within 96 -120 hours of starting ovarian stimulation with gonodotrophin . Continue upto and inculding day of ovulation injection Alternatively a single s.c. inj on day 7 of ovarian stimulation If ovarian induction is not possible on the 5th day after injection aditionaly inject 0.25mg once daily until day of ovulation

Pharmacology-
GnRH induces the production and release of LH and follicle -stimulating Hormone(FSH)
from the gonadotrophic cells at the anterior pitutary. Because of positive estradiol E2
feedback at midcycle, GnRH liberation is enhanced , resulting in LH surge. The LH surge
induces the ovulation of the dominant follicle , resumption of oocyte meiosis and
subsequently luteinization , as indicated by rising progesterone levels
