PABA +
Dapsone- dapsones antimalarial effect may be antagonised by PABA due to interference with its primary mechanism of action Procainamide
PABA inhibits procainamide - conversion to N-acetyl procainamide and it inhibits renal excetion of N-acetylprocainamide
Indication:
Effective in treatment of scleroderma
Adverse Reaction:
Anorexia Nausea Fever Rash
Contra-Indications:
Concurrent sulfonamide use (PABA inhibits the bacteriostatic activity of sulfonamides)
Safety prcautions-
Anorexia or nausea- should anorexiar nausea occur interrupt therapy until the patient is eating normally again. This avoids the possdibilty of hypoglycemia.
Hypersentivity- if hypersentivity reactions occur, discontinue the drug
Warnings- Renal functon impairment- use cautiously
Pregnancy- Safety for use during pregnancy has not been established. Use when clearly needed and when potential benefits outweigh potential hazards of the fetus.
Lactation- use only when clearly needed.
Dosages/ Overdosage Etc:
Indication-
Effective in treatment of scleroderma
Dosage
Take with food. Take tablets witrh an adeqiate amount of water to prevent GI upset. Adults- 12 g daily in 4 to 6 divided doses. As dietary supplement, take one tablet daily. Children- 1g/10 ib (4.55Kg) daily in divided doses.
Storage/stabilty- store powder in a tight light resistent container. Overdosage- Symptoms Nausea, vomiting are most common Depression of the leukocyte count and an alleged fatal case of toxic hepatitis have occured

Pharmacology/ Pharmacokinetics:
Pharmcology-
PABA is a component of of several biological systems and participates in a number of biologic processes. By increasing oxygen supply at the tissue level PABA enhances MAO activity and prevents or causes ,regression of fibrosis. These effects are speculative.
