Drug Interaction:
Alkylating Agents Include-
Mechlorethamine ,Chlorambucil, Melphalan, Ifosamide,Cyclophosphamide, Urasil Mustard, Lomustine, Carmustine, Streptozocin, Thiotepa, Busulfan, Pipobroman, Cisplatin, Carboplatin
Reduced activity of cytochrom. P-450 leads to inactivation of cyclophosphamide, procarbazine, etoposide, and nitrosoureas Interferes with adsorption of other drugs given orally.
Indication:

Adverse Reaction:
Bone marrow supression,second malignancy. Bone marrow depression, irrerversible hypoplastic anaemia Headache, dizziness,allergy, impaired fertility, hypersensitivty reaction.
Thrombocytopenia and leucopenia. Ulceration and necrosis on extrvasation,local irritation at site of injection. Intravesical instillation: haemorrhagic cystitis.
Contra-Indications:
Bone marrow depression, acute infections, hypersens.
Special precautions: Severe hepatic renal dysfunction, regular blood counts.
Dosages/ Overdosage Etc:
Indications: Adenocarcinoma of breast, ovary.
Dosage: IV admin - 0.3 to 0.4 mg/kg at 1 to 4 week intervals by rapid administration.. Storage: Store the original powder and the reconstituted solution in refrigerator at 2 to 8C. Reconstituted solution stable when refrigerated.

THIOTEPA- SYSTEMIC
1.Allergies-
Tell your doctor if you have ever had any unusual or allergic reaction to
thiotepa.
2.Pregnancy-
Tell your doctor if you are pregnant or intend to have children. There is a chance
that this medcine may cause birth defects if either the male or female is taking it
at the time of conception or it is taken during pregnancy.
Tell your doctor right away if you think you have become pregnant while receiving
thiotepa
3. Breast-feeding-
Tell your doctor if you are breast feeding or if you intend to breast feed during
treatment with this medicine. Breast feeding is not generally recommended
while you are taking it.
4.Children-
There is no specific information comparing use of thiotepa in children
5.Older adults-
Many medicines have not been studied specifically in older people. Therefore,
it may not be known whether they work exactly the same way they do in
younger adults, or if they cause different side effects or problems in older people.
Although ,there is no specific information comparing use of thiotepa in the elderly
with use in other age groups, this medicine is not expected to cause different side
effects or problems in older people.
6. Other medicines-
Tell your doctor if you are using any other medicine which may interact with
Thiotepa and cause unwanted side effects.
7. Other medical problems-
The presence of other medical problems may affect the use of Thiotepa.

Pharmacology:
Thiotepa is a cell cycle nonspecific alkylating agent. Its radiomimetic action is believed to occur through the release of ethylene radicals which disrupt the bonds of desoxyribonulceic acid (RNA) Pharmacokinetics: Urinary excretion of thiotepa and metabolites was 63%.
