Tricylic Compounds include-
Tertiary amines - Amitriptyline,Clomipramine,Doxepin,Imipramine,Trimipramine
Secondary amines - Amoxapine, Desipiramine, Nortriptyline,Portriptyline
Refer - Amitriptyline
Potentiates sedative effect of alcohol.
Antiparkinson agents antipsychotic drugs increase risk of anticholinergic effects
Reduce effect of antihypertensives,marked hyperpyrexia, convulsions & coma with MAOIs.
Potentiates hypertensive effects of sympathomimetics.
Reduced serum levels with barbiturates.
Neuroliptics inhibit metabolism.

Adverse Reaction:
Ventricular fibrillation in patients with pre-existing cardiac disease.
Agranulocytosis, thrombocytopenia.
Postural hypertension,tachycardia,cardiac arrhythmias, conduction disturbances.
Dry mouth,blurring of vision, constipation, urinary retension, fatigue, weakness, dizziness.
Tremors, headache, confusion and delirium in elderly, weight gain, sexual disturbances.
Contra-Indications:
Recent MI,heart block or other cardiac arrhythmias
.Mania, severe liver disease.
Special precautions:
May cause blood dyscrasias,hepatitis and peripheral neuropathy & hemolytic anaemia.
Regular opthalmic examination in long term therapy.

Indications:
Relief of symptms of depression.
Dosage:
Amoxipine is not recommended for patients below 16 years old. Adults- Initially 50mg 2 or 3 times daily, increase to 100mg 2 or 3 times daily by the end of the first week .
The total daily dose not to exceed 300mg.
Elderly- initially 25mg 2 or 3 times a day. Increase to 50mg 2 or 3 times by the end of first week. 100 to 150mg/day may be adequate for elderly patients..
