Amantadine +
Triamterene
Amantadine plasma level incr & urine excertion decreased with risk of adverse effects
Anticholinergic drugs
Reduce the dose of anticolinergic drugs or of amantadine if atropine-like effects appear when these drugs are used concurrently.
Decreased urinary excretion of amantadine with subsequent increased plasma concentration occured when hydrochlorthiazide+ triamterene was administered concurrently with amatadine
Indication:
Parkinsons disease/syndrome and drug induced extrapyrimidal reactions.
Adverse Reaction:
Nausea,dizziness,light headedness,
insomnia. depression, anxiety, irritability,
hallucination, confusion, less frequently noticed.
Overdose symptoms- Nausea,vomiting,anorexia and CNS effects, Tremors,ataxia,blurred vision,lethargy,depression.
CNS toxicity- treat with IV physostigmine 1 to 2mg administered every 1 to 2hrs in adults or 0.5mg at 5 to 10 minutes intervals upto a max of 2mg/hour in children.
Contra-Indications:
Hypersens to amantadine,
pregnancy,lactation.
epilepsy, severe renal impairment and gastric ulceration.
Special precautions:
May cause CNS effects or blurred vision,
Observe caution while driving or performing other tasks requiring alertness.
Do not discontinue abruptly.
Few patients with Parkinsons disease experienced parkinsonian crisis( sudden marked clinical deterioation)when this medication was stopped suddenly.
Dosages/ Overdosage Etc:
Indications:
Parkinsons disease/syndrome and drug induced extrapyrimidal reactions.
Dosage:
100mg twice a day when used alone. Onset of action is within 48hrs.
Initial dose is 100mg/day for patients with serious associated medical illness after one to several weeks at 100mg/day increase to 100mg twice /day.

Patient Information:
Amantadine
1. Individual allergy - Tell your doctor if you have had any unusual or allergic reaction to amantadine. Also tell your doctor, if you are allergic to any other substances such as foods, preservatives etc.
2. Pregnancy - Studies have not been done in humans. Some animal studies have shown that amantadine is harmful to the fetus. Follow your doctors advise.
3. Breast feeding - Amantadine passes into breast milk. Effects of amantadine in newborn babies and infants are not known. Tell your doctor and follow his advice.
4. Children - Medicine has been tested in children over 1 year of age and has not been shown to cause different effects or problems in these children than it does in adults. Follow the advice of your doctor.
5. Other medicines - Certain medicines should not be taken together at all. Tell your doctior if you are taking other prescription medicines, especially- amphetamines, caffeine, cocaine, medicine for asthma, medicine for colds, sinus problem, or hay fever, antocholinergics for stomach spasms or cramps.
6. Other medical problems - Tell your doctor if have other medical problems- eczema, epilepsy orseizures,heart disease or circulation problems, swelling of the feet and ankles. kidney disease, mental or emotional illness.
Tell your doctor, if you are prone to flu.
7. Dose - Take this medicine exactly as directed by the doctor. Do not miss any dose and or do not take more than ordered by your doctor.
8. Missed dose - If you miss your dose, take it as soon as possible. However, if it is almost time for your next dose, skip the missed dose and go back to your regular dosing schedule. Do not double doses.
9. Storage - Keep out of reach of children. Store away from heat and direct sunlight. Do not store in the bathroom or near the kitchen sink. Moisture and heat may cause the medicine to break down. Do not keep outdated medicenes, Keep discarded medicines out of reach of children.

The exact mechanism if action is not well known,but amantadine is thought to release dopamine from intact dopaminergic terminals that remain in the substantial nigra of parkinson patients.

Pregnancy and lactation:
Contraindicated for use during pregnancy and lactation.
